Research subject for a potential FP6 project

Cell surface antigens of leukocytes (CD antigens/molecules) are important for immune reactions of organism. They are selectively present on some subpopulations and functional types of leukocytes. CD antigens participate in immune reaction as receptors for cell communication (e.g. adherence molecules, antigen recognising receptors). CD molecules are commonly detected with monoclonal antibodies (mAbs) produced by hybridoma technique. In the last decade the wide palette of monoclonal antibodies has been prepared which recognise of CD antigens on human cells. Much less monoclonal antibodies are available specific for typing of domestic animal cells. Therefore the production of new mAbs specific for animal cells is desirable.The main aim of the proposal is the construction of hybridomas for production of monoclonal antibodies recognising the cell surface markers of leukocytes of domestic animals. The proposal comprises the production of hybridomas, biochemical, cytological, histochemical analyses of cell markers with prepared antibodies to determine the specificity and antigen affinity of prepared antibodies. The well defined hybridomas are attractive for their commercialisation and use, for example, to highlight their capacity to immunoprecipitate and/or react with tissue sections. The commercialisation is possible via companies involved in selling of immunodiagnostic reagents. The main field of the application is the veterinary immunodiagnostic reagents used in the diagnosis of primary and secondary immunodeficiencies of domestic animals. Furthermore, monoclonal antibodies prepared in the project could be used in the biomedical research. The participation in the project could be attractive for the laboratories in Europe which have experience with the production of monoclonal antibodies. The international cooperation is highly desirable to concentrate the experimental skill and knowledge in this field.
